| 背景說明: | 生長分化因子5(GDF-5，BMP-14)是轉化生長因子β超家族蛋白家族中的一員。人GDF-5、-6和-7是BMP家族的一個定義子群.GDF-5是一種由354個氨基酸(AA)N端前體和120 aA C端成熟肽組成的同源二聚體前體蛋白。成熟人GDF-5與成熟小鼠和大鼠GDF-5具有99%的AA序列同源性.GDF-5信號轉導途徑是由1型(BMPR-IB)和Ⅱ型(BMPR-IIor Activin RII)絲氨酸/蘇氨酸激酶受體(Smad 1、5和8)組成的雜二聚體復合物介導的。GDF-5參與了肢體發生、軟骨發育、關節形成、骨形態發生、細胞存活和神經發生等多種發育過程。抑制GDF-5的表達或改變其信號通路可促進骨關節炎的發生. |
