pCold-SUMO質粒 pColds-SUMO載體簡介
原核表達載體(pCold-SUMO)是在 pCold 載體基礎上改造而成,該載體啟動子(CS Promoter)來源于南極嗜冷細菌,在低溫下(15 度)才能啟動蛋白的表達 。低溫下細菌生長緩慢,使得蛋白合成速度減慢,從而最大限度的提高了蛋白正確折疊的幾率,提高了蛋白可溶性表達,增強了活性蛋白的表達比率。該表 達系統所包含的 SUMO tag可以極大地提高小分子量蛋白的表達量,而且進一步提高了蛋白的可溶表達幾率。同時,試劑盒配備的SUMO Protease(含 6×His標簽)可特異性去除 SUMO tag,從而得到不含任何標簽的重組蛋白。TEE信號肽可增強冷啟動子調控下目的蛋白的高表達。BL21(DE3)Chaprone E.coli 細菌中含有分子伴侶蛋白質粒,其表達產物可協助重組蛋白正確折疊形成可溶活性蛋白。 分子伴侶蛋白質粒為氯霉素抗性(chloramphenicol,Cmr)的表達受控于四環素(tetR)操縱子。
