pRSET C質粒 pRSETC載體簡介
pRSET C載體是從pUC的載體衍生而來,載體設計的目的是為了在大腸桿菌中高水平的表達和純化目的克隆基因。pRSETC載體上的T7啟動子使得載體高水平表達克隆的基因序列成為可能。
此外,pRSET-C載體的DNA插入片段位于下游側,并能夠上游的蛋白編碼框相一致,上游的編碼序列表達了一個N-端融合的肽。該序列包括一個ATG翻譯起始密 碼子,一個6 X His標簽,一個來源于噬菌體T7基因10的轉錄穩定序列,Xpress抗原表位,以及一個腸激酶裂解識別序列。
His 標簽允許簡單的Ni柱蛋白純化。腸激酶識別位點位于His標簽和重組蛋白之間,可以隨后方便的去除重組蛋白的N-端的融合肽。
