pCold TF質粒 pColdTF載體簡介
Takara的pCold TF載體是一種融合表達可溶性的標簽"觸發因子(Trigger Factor,TF)伴侶"的冷休克載體。"觸發因子"(48KD)是原核核糖體相關的分子伴侶蛋白(48 kDa),有利于新表達的多肽的共翻譯折疊。正因為它來源于大腸桿菌,保證了觸發因子能夠在大腸桿菌表達系統中高表達。 pCold TF載體由CSPA啟動子及其下游的序列組成,其下游序列主要包括包括5'非翻譯區(5'非編碼區),翻譯增強元件(TEE),His標簽序列,多克隆位點(MCS)。Lac操縱子插入在CSPA啟動子下游,以確保嚴格的表達調控。此外,HRV 3C蛋白酶,凝血酶,和Factor Xa識別位點位于“Trigger Factor”和多克隆位點(MCS)之間,以便表達的融合蛋白上將標簽去除。
大多數大腸桿菌菌株都可以可以作為本載體的表達宿主。pCold TF載體提供冷休克誘導的方法高水平表達目的蛋白,再加上觸發因子(伴侶)的表達來提高正確的蛋白質折疊,從而實現高效可溶性表達其他系統中難以對付的蛋白。
