pUG66載體是大腸桿菌-酵母穿梭載體,在大腸桿菌中抗性是氨芐。pUG66載體質粒含有來自Tn5載體質粒的Ble基因,使得載體質粒在酵母菌中呈現出博來霉素抗性。 pUG66載體在篩選基因Ble兩側含有loxp基因,通過表達cre重組酶,可以利用loxp基因將Ble s篩選基因刪除。 pUG66載體可以作為酵母基因敲除的一個質粒PCR模板。代表性的PCR片段的整合插入宿主菌,使得基因敲除突變子能夠進行順利篩選。 |
