酚紅葡萄糖肉湯培養基（Phenol Red Glucose Broth）是一種常用的基于微生物葡萄糖發酵能力的 鑒別性液體培養基。
Phenol Red Glucose Broth的工作原理：
細菌的鑒別歸根結底是根據其能夠分解利用哪些化合物為依據的，即能夠產哪些酶。酚紅葡萄糖肉湯 培養基的鑒別依據是微生物發酵利用葡萄糖的能力。葡萄糖發酵型微生物在水解利用葡萄糖過程中，產生酸性副產物，使培養基的pH下降 。有些葡萄糖發酵型微生物產酸也產氣（主要為二氧化碳）。非葡萄糖發酵型微生物以蛋白胨為碳源，在有氧條件下，利用氨酸酸脫氨基 作用，獲得碳水化合物，產生堿性副產物NH3。或者通過氨基酸異化作用獲得碳源，不產堿性副產物。
培養基配方中的葡萄糖為葡萄糖發酵型微生物提供直接碳源。牛肉提取物提供生長因子。蛋白胨作為主要氮源，也是非葡萄糖發酵型微生 物的碳源。酚紅是酸堿指示劑，變色范圍為pH6.8(黃色）~pH8.4(紫紅色)。酚紅溶液在pH<6.8時為黃色，pH中性時為紅色，pH>8.4時則 顯紫紅色。當培養基顏色由紅色變黃色時，說明葡萄糖被發酵產酸。如果培養基顏色不變或紅色加深，說明該微生物葡萄糖發酵陰性。葡 萄糖發酵試驗是在發酵管中進行的，發酵時若有氣體生成，會被滯留在倒置的德拉姆發酵管中，形成氣泡。

配制方法：
1. 稱取脫水培養基21.0g，用1L去離子水溶解。
2. 分裝到含有德拉姆發酵管的試管中。
3. 121°C高溫蒸汽滅菌15min。
滅菌后的液體培養基置于2-8ºC保存，蓋緊管蓋。避免光線直射。
實驗方法——葡萄糖發酵試驗（Glucose Fermentation Test）
1. Select the phenol red Glucose broth medium.
2. Start your Bunsen burner.
3. Select the inoculating loop tool.
4. Flame your inoculating loop to sterilize it.
5. Remove the caps from your test tubes.
6. Flame the mouths of your test tubes.
7. Use the sterile inoculating tool to pick up an inoculum from the culture tube of the unknown bacterium.
8. Immediately transfer the inoculum into the fresh, sterile medium.
9. Flame the mouths of your tubes once again.
10. Replace the caps on the test tubes.
11. Re-flame the inoculating tool.
12. Place the inoculated tube into the 35-37 C incubator.
13. Incubate for the appropriate length of time. For this test, 24 hours is sufficient for determining the result.
14. Retrieve desired incubated culture from the incubator.
15. Observe test result. If the test was followed as described above, the culture will have changed to yellow in the presence of acids (indicating a positive test) or magenta or hot pink in the presence of bases/alkali (indicating a negative test).
16. Record test result.
17. Dispose of the culture.
實驗結果與分析
所有的腸桿菌科細菌在該培養基中都能良好生長，發酵產生的酸除了使pH發生改變外，其中的丁酸還會給培養物散發強烈的臭味。
